Effect of diphosphonates on adenosine 3':5'-cyclic monophosphate in mouse calvaria after stimulation by parathyroid hormone in vitro.

چکیده
1. The diphosphonates, disodium ethane-1-hydroxy-1,1-diphosphonate (EHDP) and disodium dichloromethylene diphosphonate (Cl2MDP), inhibit bone resorption in animals and in explanted bone in tissue culture. The possibility that these effects might be due to inhibition of skeletal adenylate cyclase has been studied. 2. EHDP and Cl2MDP, added for 30 min to the incubation medium at concentrations known to inhibit bone resorption, had no effect on basal content of adenosine 3':5'-cyclic monophosphate (cyclic AMP) of mouse calvaria incubated in vitro, nor did they inhibit the rise in cyclic AMP induced by bovine parathyroid hormone. 3. Pretreatment of mice for 3 days with Cl2MDP also had no effect on cyclic AMP under basal conditions or after incubation of explanted calvaria with parathyroid hormone in vitro. EHDP under similar conditions slightly inhibited the increase induced by parathyroid hormone but had no effect on basal concentrations of cyclic AMP. 4. It is suggested that the inhibition of adenylate cyclase is not an essential feature of the reduction of bone resorption by diphosphonates, which may act by direct inhibitory effects on the dissolution of hydroxyapatite and perhaps by other unidentified effects on bone cells. Key words: adenosine 3':5'-cyclic monophosphate, bone, dichloromethylene diphosphonate, diphosphonates, ethane-1-hydroxy-1,1-diphosphonate, parathyroid hormone.

The effect of parathyroid hormone on the concentration of adenosine 3',5'-monophosphate in skeletal tissue in vitro.
Adenosine 3’,5’-monophosphate (3’,5’-AMP) was assayed in calvaria isolated from fetal rats at term. A marked increase in 3’,5’-AMP was detected in vitro within 1 min after adding purified parathyroid hormone to calvaria incubated in Krebs-Ringer-bicarbonate or Krebs-Ringer-Tris buffer. A maximal effect was detected after 5 min of incubation with added parathyroid hormone. The lowest concentrati...
متن کاملThe Effect of Parathyroid Hormone and Thyrocalcitonin on the Accumulation of Cyclic Adenosine 3’: S-Monophosphate in Freshly Isolated Bone Cells*
Bone cells were isolated from embryonic rat calvaria by a modtied method designed to give larger yields and undamaged membranes. Essentially it consists of perfusing bone chips with fresh collagenase, collecting the cells continuously in the efiluent, and washing off the collagenase. Cyclic adenosine 3’:5’-monophosphate (CAMP) accumulation in the freshly isolated cells was measured by equilibra...

متن کاملCardiotonic agent milrinone stimulates resorption in rodent bone organ culture.
The cardiotonic agent amrinone inhibits bone resorption in vitro. Milrinone, an amrinone analog, is a more potent cardiotonic agent with lower toxicity. In contrast to amrinone, milrinone stimulated resorption in cultures of neonatal mouse calvaria and fetal rat limb bones. Threshold doses were 0.1 microM in calvaria and 0.1 mM in limb bones; maximal stimulation occurred in calvaria at 0.1 mM. ...
متن کاملRainbow trout hypocalcin stimulates bone resorption in embryonic mouse calvaria in vitro in a PTH-like fashion.
Hypocalcin, the major hormone with hypocalcaemic action in fish, was isolated from trout corpuscles of Stannius (SCs). The bioactivity of hypocalcin was assessed in a parathyroid hormone (PTH) bioassay involving bone resorption in embryonic mouse calvaria. Calcium and phosphate release and lactate production were stimulated in a dose-dependent manner by hypocalcin. On a molar basis about equal ...
